Subtraction cDNA-library

Introduction
Subtractive cDNA cloning is a powerful technique which allows the selective amplification of genes which are differentially transcribed, for example in activated but not in resting cells or in one tissue but not in another. With the help of substrated cDNA libraries these genes can be isolated and identified.

After working through this topic you should be able to
explain the construction of a PCR-based substracted cDNA library.

The method
The construction of a subtracted cDNA library is described in the milestone „AID, an enzyme essential for class switching and somatic hypermutation“.
