Introduction
Limiting dilution assays may underestimate the number of specific T cells because this technique cannot detect cells with
low or none proliferative potential. Yet, staining specific T cells with their physiological
ligand fails because soluble peptide-MHC-complexes dissociate very rapidly form the T cell receptor.
